Our recently obtained spectral flow cytometry analyzer Sony ID7000E harbors 6 lasers (320nm, 355nm, 405nm, 488nm, 561nm, 637nm) with full-spectral detectors for spectral unmixing. The 6 lasers enable adequate differentiation of fluorophores and fluorescent proteins with similar emission spectra and improves autofluorescence detection. The ID7000E furthermore constitutes an autosampler for automated acquisition of 24 samples in 5ml tubes as well as 96-well and 384-well plates and a constant cooling system that can be set to 4°C. This analyzer is located within the BSL2 area.

An additional ID7000 analyzer with 5 lasers (355nm, 405nm, 488nm, 561nm, 637nm) and a conventional flow cytometer (BD LSRFortessa) are located in CECAD (BSL1). If you’re interested in using these devices, please visit the Flow Cytometry Core website or contact ccf-flow@uk-koeln.de.

Our newly acquired spectral flow cytometry sorter Sony FP7000 features 6 lasers (320nm, 355nm, 405nm, 488nm, 561nm, 637nm) with full-spectral detectors for spectral unmixing. The 6 lasers enable adequate differentiation of fluorophores and fluorescent proteins with similar emission spectra and improve autofluorescence detection. The FP7000 furthermore enables the simultaneous sorting of up to 6 different cell populations from a single sample. Index sorting allows traceability of single cells sorted into plates. The FP7000 constitutes different nozzle sizes (70µm, 100µm, and 130µm) to ensure gentle sorting of cells with different diameters and is capable of sorting into 1.5ml to 50ml tubes as well as 6-well to 384-well plates. A constant temperature control system can be set to 4°C (sample and collection tubes) and 37°C (sample tube).

The BD FACSAria Fusion is a conventional cell sorter and equipped with 4 lasers (405nm, 488nm, 561nm, 640nm) and 17 filters, which facilitates robust cell sorting into 96well-plates and different tubes and is located in the BSL2 area.

A powerful workstation for flow cytometry data analysis is available in the CMMC (R. 5.013) and can be booked via the FCC. Furthermore, software licenses for analysis software (i.e. FlowJo) are available on the workstation as well as for your own computer.

Within the Flow Core, the gentleMACS Octo tissue dissociator for automated tissue dissociation, homogenization, and perfusion is available in the CMMC (R. 5.013).
